Ampakines do not increase the amount of glutamate released. They bind to a site on the AMPA receptor and, when glutamate binds to the channel, slow the closure of that channel. This means that there will be a prolonged current flow of Na+ ions in the cell, making it more likely that a strong depolarization will occur.
