History of Adrafinil

Adrafinil was first developed by researchers at the French pharmaceutical company Lafon Laboratories. In 1974, Michel Jouvet and another scientist created a series of benzhydryl sulfinyl compounds including Adrafinil, while researching new analgesics.
Adrafinil was found to not have pain-relieving effects, but rather belongs to a class of compounds known as eugeroics, or wakefulness promoting compounds. Adrafinil has stimulant like effects, but is not related structurally or in its mechanism of action to traditional amphetamine based stimulants.
During testing to determine what Adrafinil’s mechanism of action was, Modafinil was discovered in 1976. When ingested, Adrafinil is transported to the liver, where it is broken down into its component parts. These include the inactive modafinilic acid, and the active Modafinil, which has been determined to cause Adrafinil’s effects.
Both Adrafinil and later Modafinil were seen in animal trials to have stimulating effects, causing increased locomotion and alertness. Because of these effects, Adrafinil was first tested in human trials as an experimental treatment for narcolepsy. Narcolepsy is a sleep disorder that causes excessive daytime sleepiness and random bouts of sleeping during the day.
In various double-blind placebo-controlled research studies, compared to a placebo, Adrafinil was seen to significantly increase wakefulness and reduce excessive daytime sleepiness. In 1986 Adrafinil was approved in France as a treatment for narcolepsy. Later Modafinil was approved as well in France in 1994, and in the US in 1998.
Lafon made brand name version of both Adrafinil and Modafinil, known as Olmifon and Provigil respectively. In 2001, Lafon was bought by Cephalon Inc, an American pharmaceutical company. Cephalon continued production of Olmifon and Provigil.
In 2011, Cephalon was acquired by Teva Pharmaceutical Industries. That same year, production of Olmifon was ceased. Modafinil is the stronger and faster-acting compound compared to Adrafinil, and is the one that has been studied more extensively and used in medicine.
