Adrafinil was first developed by a couple of scientists at Lafon Laboratories, a French pharmaceutical company, in 1974. Experimental medicine professor Michel Jouvet and another scientist were researching new analgesics when they developed a group of benzhydryl sulfinyl compounds, one of which came to be known as Adrafinil.
It was determined that Adrafinil did not have any pain-relieving capabilities, but instead belonged in a class of compounds known as eugeroics. Eugeroics are wakefulness promoting compounds that have stimulant-like effects, but do not work in the body the same way as traditional amphetamine based stimulants to cause effects.
During research to determine Adrafinil’s mechanism of action in the body, the compound Modafinil was discovered. When it is ingested, Adrafinil is transported to the liver, where it is metabolized and broken down into its component parts, namely the inactive modafinilic acid and Modafinil.
Modafinil has been determined to be the primary active compound in Adrafinil. This makes Adrafinil a prodrug, a compound that is largely inactive on its own that is converted into an active compound in the body.
Both Adrafinil (through the action of the Modafinil it is converted into) and Modafinil on its own have been observed to have stimulating, alertness promoting effects in animal trials. This led to the first human trials using Adrafinil as a treatment for narcolepsy, a sleep disorder that causes excessive daytime sleepiness and random sleeping bouts during the day.
Multiple double-blind placebo-controlled research trials first using Adrafinil and later using Modafinil alone have found that compared to a placebo the compounds significantly improved wakefulness and reduced excessive sleepiness.
